Development of a Biodegradable Microcarrier for the Cultivation of Human Adipose Stem Cells (hASCs) with a Defined Xeno- and Serum-Free Medium

Stirred single-use bioreactors in combination with microcarriers (MCs) have established themselves as a technology that has the potential to meet demands of current and future cell therapeutic markets. However, most published processes been performed using fetal bovine serum (FBS) containing culture medium non-biocompatible MCs. This approach two significant drawbacks: firstly, inevitable risks associated use FBS for clinical applications; secondly, MCs be removed from suspension before implantation, requiring step causes loss viable cells adds further costs complications. study aimed develop new platform based on chemically defined xeno- serum-free biodegradable MC can support growth human adipose stem (hASCs) while still preserving their undifferentiated status. A specific components manufacturing parameters resulted prototype, called “BR44”, which delivered desired functionality. BR44 allows hASCs stick its surface grow (UrSuppe). Although cells’ expansion rate was not high commercial non-biodegradable standard MC, those cultured maintained better status both static dynamic conditions than traditional 2D surfaces.
